What Stands Out
Product Details
- 10X TBE Buffer (Tris-borate-EDTA) is the most commonly used buffer for DNA and RNA polyacrylamide gel electrophoresis. It is also routinely used for DNA automated sequencing gel. TBE can also be used for agarose gels, but is not recommended for preparative gels for recovery of nucleic acids. Since borate in TBE buffer is a strong inhibitor for many enzymes, TAE buffer is recommended when looking at enzymatic applications for the DNA sample.
- Applications: Electrophoresis of nucleic acids in agarose and polyacrylamide gels
- Used both as a running buffer and as a gel preparation buffer
- Filtered through a 0.45 ?m membrane
- Recommended for electrophoresis of RNA and DNA fragments smaller than 1500 bp

Product Description
Customer Questions & Answers
-
Question:
What is 10x TBE Buffer and its primary use?
Answer: 10x TBE Buffer, comprising Tris base, boric acid, and EDTA, is primarily used in molecular biology for running agarose gels, a crucial step in DNA analysis. This buffer maintains a stable pH during electrophoresis, ensuring optimal separation and visualization of nucleic acids. Laboratories commonly use TBE Buffer for various applications, including DNA purification, restriction digestion, and plasmid analysis, making it an essential reagent in genetic research and biotechnology. -
Question:
How should I prepare 10x TBE Buffer for use in gel electrophoresis?
Answer: To prepare 10x TBE Buffer for gel electrophoresis, you must dilute it with distilled water to achieve a 1x concentration. The general guideline is to mix 100 mL of 10x TBE with 900 mL of distilled water. This dilution is critical as it provides the necessary ionic strength for effective DNA separation. For routine laboratory processes, maintaining a prepared stock of diluted 1x TBE Buffer simplifies preparation for gel casting and running experiments. -
Question:
What is the difference between 1x TBE Buffer and 10x TBE Buffer?
Answer: The primary difference between 1x and 10x TBE Buffer lies in their concentrations. The 10x TBE Buffer is a concentrated stock solution that must be diluted to 1x prior to use for any applications. The 10x concentration ensures that the buffer can be easily stored and transported while retaining its stability. Laboratories often utilize the 10x form for convenience, allowing for quick preparation of larger volumes of the diluted buffer as needed for various experiments. -
Question:
Can I use 10x TBE Buffer for RNA gel electrophoresis?
Answer: Yes, 10x TBE Buffer can be used for RNA gel electrophoresis, although it is primarily favored for DNA separations. Its properties help maintain the integrity and charge of RNA molecules, facilitating effective resolution during electrophoresis. However, researchers often recommend using 1x MOPS or 1x TAE buffers for RNA, as they might offer better results in specific applications. Consider the requirements of your experiment to choose the most suitable buffer for optimal outcomes. -
Question:
What precautions should I take when handling 10x TBE Buffer?
Answer: When handling 10x TBE Buffer, it’s important to use appropriate personal protective equipment, such as gloves and goggles, as the boric acid component can be irritating. Always work in a well-ventilated area and avoid skin contact. Proper storage away from heat and light is recommended to maintain buffer integrity. These precautions ensure safety during lab procedures while minimizing contamination risk, which is crucial for reliable experimental results. -
Question:
Can I store diluted 1x TBE Buffer for future use?
Answer: While diluted 1x TBE Buffer can be stored, it's best used fresh for optimal results. If you need to store it, keep it in a clean, airtight container at 4°C and aim to consume it within a few weeks. Over time, the pH balance can change, affecting performance in gel electrophoresis. If precision is essential for your experiments, consider preparing directly from 10x stock shortly before use to ensure the best results. -
Question:
Is 10x TBE Buffer suitable for southern blotting techniques?
Answer: Yes, 10x TBE Buffer is suitable for Southern blotting, a technique for detecting specific DNA sequences within a complex mixture. Its ability to maintain stable pH during the electrophoretic separation of DNA ensures effective transfer onto membranes for hybridization processes. Laboratories routinely use this buffer for its effectiveness in preserving DNA's integrity while preparing it for blotting and subsequent analysis. -
Question:
What adjustments should I make when using 10x TBE Buffer with different agarose concentrations?
Answer: When using 10x TBE Buffer with various agarose concentrations, you typically don't need to adjust the buffer itself, but the concentration of agarose will affect the resolution of the DNA or RNA. For lower DNA fragment sizes, use a higher percentage of agarose gel, while larger fragments require lower agarose percentages. Always ensure that regardless of the concentration used, the 10x TBE is adequately diluted to 1x to maintain consistent buffer performance during electrophoresis. -
Question:
How does TBE Buffer compare to TAE Buffer in electrophoresis?
Answer: TBE Buffer and TAE Buffer are both commonly used for electrophoresis, but they have distinct properties. TBE Buffer offers superior buffering capacity and is particularly effective for long runs, as it allows for better resolution of DNA fragments. Conversely, TAE Buffer is more suitable for applications involving enzymatic reactions, as it does not inhibit restriction enzyme activity. Depending on your specific application, choose TBE for better separation and TAE when enzyme activity is critical. -
Question:
